Overview
SNAP-8 (acetyl octapeptide-3) is an eight-amino acid synthetic cosmetic peptide designed as an enhanced analogue of Argireline. It mimics the N-terminal sequence of SNAP-25, a protein in the SNARE complex involved in neurotransmitter vesicle fusion at neuromuscular junctions. Through competitive interference with SNARE complex assembly, SNAP-8 is studied for its potential to moderate facial muscle contraction intensity and the resulting appearance of expression lines. Research Background
SNAP-8 was developed as an extension of Argireline research, with two additional amino acids providing closer structural mimicry of the SNAP-25 N-terminus. In vitro and topical application studies have reported greater inhibition of catecholamine release and more pronounced reductions in expression wrinkle depth compared to Argireline at equivalent concentrations. Independent peer-reviewed research is more limited than for systemic research peptides.
Studied Mechanisms
SNAP-8 competes with SNAP-25 for incorporation into the SNARE complex at neuromuscular junctions, reducing the efficiency of acetylcholine vesicle fusion. This moderates neuromuscular transmission intensity without blocking it completely. The result is reduced contraction amplitude in repeatedly contracting facial muscles. Research has also studied transdermal delivery formulations to address the penetration challenge of topical peptide delivery.
